LATE-PCR for LoC Molecular Diagnostics Devices and Its Application to the Sensitive Detection of SARS-CoV-2

The emergence of the novel coronavirus, SARS-CoV-2, has highlighted need for rapid, accurate, and point-of-care diagnostic testing. Lab-on-a-Chip (LoC) devices offer possibility to run such tests at a low cost, while same time permitting multiplexed detection several viruses when coupled with microarray amplified products. Herein, we report development protocol qualitative through design appropriate primers that target evolutionary conserved regions virus. proposed relies on an improved version asymmetric RT-PCR, linear-after-the-exponential (LATE)-PCR uses are deliberately designed use unequal concentrations. As result, LATE-PCR exhibits similar efficiency symmetric PCR, promoting accumulation single-stranded products can subsequently hybridize single-strand DNA probe-spotted microarray. performance developed was compared validated artificial viral RNA nasopharyngeal swab samples from real patients. Furthermore, in order illustrate its potential integration into biosensor platform, amplicons were allowed probes covalently immobilized onto commercially available functionalized glass, without heat denaturation.
